Summary of Postdoctoral Fellowship:

Perceptual discriminations are among the most important decision making processes. One of the most prominent questions in systems neuroscience is how the perception of a given stimulus is encoded in the brain and how this translates to a learned decision that is followed by a motor response. Using the repertoire of experimental techniques available in our lab, such as two-photon and three-photon imaging, multi-electrode extracellular recordings, patch-clamp recordings, holography based optogenetics and psychophysics in awake behaving animals, our goal is to characterize in detail single-cell and neural network activity in the rodent cortex during the learning process of a decision making task. The postdoc will be able to choose among several projects that will use the cutting-edge infrastructure of the lab: Imaging bi-hemispheric interactions during decision making using a novel dual-head two-photon microscope. Deep imaging of prefrontal cortex activity during decision making using three-photon excitation. Holographic stimulation of neurons to probe cortical coding schemes relevant for perception (see e.g. Musall et al. 2014, Nature Neuroscience).

The brain is very hard to cure. Drugs have a hard time crossing the blood-brain barrier and if they do – the side effects of their activity in such a fragile environment may be disastrous. Our lab strives to describe and uncover an alternative approach to brain treatment. 11. Three-year Postdoctoral Position at Institut Pasteur, Paris, France (Prokaryotic Cell Biology, Evolution)

The Archaea have recently come to center stage, not only because of their key role in the origin of eukaryotes (1), but also as important yet neglected residents of the human microbiome (2). We have recently developed the first experimental model of a host-associated archaeon, Methanobrevibacter smithii, the main species of methanogens from the human gut. We have demonstrated that M. smithii is an ovococcoid that divides binarily using a homologue of FtsZ and its anchor SepF (3). Interestingly, while most archaea contain a proteinaceous S-layer as major component of the cell envelope, M. smithii and its close relatives possess a cell wall made of pseudopeptidoglycan (pPG). It structurally resembles its bacterial counterpart, but is chemically distinct, indicating an independent evolutionary origin. How these archaea “reinvented” a pPG cell wall, and what were the consequences at the cellular level are fascinating open questions. In fact, the archaeal machineries involved in cell growth and division in the presence of a pPG cell wall remain largely unknown (4).
