A rise in early-onset (age <40) type 2 diabetes has raised concerns due to its aggressive nature and poor prognosis. This project will address critical knowledge gaps regarding the causes and consequences of early-onset type 2 diabetes. It is based on a cohort consisting of the entire Swedish population with linkages between relatives and information from nationwide registers covering a 30-year time span together with biobank data for a subset. The project combines traditional cohort designs with family-based designs to minimize confounding and for familial aggregation and heritability studies.

Summary of Postdoctoral Fellowship:
The postdoctoral fellow will determine how disease-relevant long non-coding RNAs (lncRNAs) regulate adipocyte insulin sensitivity and in turn metabolic health. This will be achieved by investigating human white adipose tissue (WAT) biopsies taken before and after systemic insulin infusion and couple these findings with state-of-the-art cellular models and in-house developed molecular approaches. This project aims to understand how lncRNAs contribute to the insulin response in adipocytes and discover novel targets that can be approached therapeutically.

The Schultzberg lab addresses fundamental questions in neuroinflammation with the aim to find new treatment targets in Alzheimer’s disease. Our studies build upon in vivo and in vitro models as well as on analysis of human clinical samples, in order to elucidate the molecular and cellular mechanisms for resolving neuroinflammation and regaining homeostasis in the brain. Resolution of inflammation in the brain is a relatively new research field and our studies aim to provide knowledge on the signals for resolving inflammation in the brain and whether stimulation or modulation of these is beneficial for neuronal functioning and eventually as a potential target for the treatment of patients.

The Baranello Lab – Drawing from our expertise in the fields of topoisomerase and cancer biology, we aim to answer fundamental questions about how transcription, replication and DNA topology are functionally coordinated in human cells. We use state-of-the-art approaches to assess transcriptomic, epigenomic and replisomic profiles, as well as advanced microscopy and drug screens to understand how topoisomerase regulation affects fundamental nuclear processes (key lab references: Baranello et al. Cell 2016; Wiegard et al. Molecular Cell 2021; Das et al. Molecular Cell 2022; Cameron et al. Science Advances 2023). The overall aim is to reveal novel cancer therapy targets and improved strategies for personalized treatment.

The Larsson lab is a research group within the Department of Oncology-Pathology at Karolinska Institutet and is situated at the Science For Life Laboratory at the Solna Campus. The focus of the group is on achieving transcriptome-wide and detailed molecular understanding of how mRNA translation is dysregulated in disease. The lab has expertise in development of state-of-the-art methods for large-scale wet-lab studies of mRNA translation as well as development of computational tools to analyse the generated data. The post-doc will use advanced molecular biology approaches to generate large data sets quantifying various aspects of the mRNA translation process, which will be analysed to gain detailed mechanistic insights.
