The aim of the overall research project is to develop an empirically informed philosophical framework for addressing the conservationist’s dilemma. This dilemma arises from the fact that, in their efforts protect (critically) endangered species, conservation policy makers can be confronted with disputes among biological taxonomists about how to individuate those species in the first place. On the one hand, species conservation programs cannot wait for the resolution of such disputes; on the other hand, these programs risk being misdirected if they are rooted in flawed taxonomic perspectives. The projects are part of the research project “PlantsGoImmune”, lead by Professor Hans Thordal-Christensen and financed by Novo Nordisk Fonden. Plants have a highly complex immune system where recognition of pathogens activates signalling cascades to trigger immune responses. Using the powdery mildew fungus, we have uncovered that membrane trafficking processes in the plant cell are essential for building immune structures at the site of invasion. Alternatively, the attacked plant cell can undergo a programmed cell death reaction that stops this pathogen. Meanwhile, pathogens secrete effector proteins that target and counteract these immune responses.

The glaucoma drug pilocarpine is produced by jaborandi plants, which are native to the Amazon rainforest. In this collaborative project, the Geu-Flores laboratory at the University of Copenhagen has joined forces with the group of Guilherme Oliveira at the Vale Institute of Technology (Belém, Brazil) to elucidate the biosynthesis pathway of pilocarpine in jaborandi. The elucidation of this fully unknown pathway will pave the way for the biotechnological production of pilocarpine and relieve the pressure on wild jaborandi, thus contributing to the preservation of biodiversity in the “Planet’s lungs”.

11. PhD Fellowship in “Protective Mechanisms of Selenocyanate in Inflammatory Disease
Summary of Doctoral Project:
Chronic inflammation is a repetitive cycle of immune cell infiltration and activation that underlies the development of many diseases, particularly atherosclerosis. Activated immune cells release the enzyme myeloperoxidase, which catalyses the reaction of hydrogen peroxide with chloride ions to form the potent oxidant hypochlorous acid (HOCl, the active component in bleach). This oxidant plays a critical role in the immune system by killing microbes to combat infection. However, excessive production of HOCl in chronic inflammation causes extensive, irreversible host tissue damage, which results in disease. As a result, there is widespread interest in the development of strategies to combat HOCl-induced damage in a number of chronic disease settings like atherosclerosis. This project will explore the use of selenocyanate as a means to prevent the tissue damage seen in chronic inflammatory pathologies without compromising innate immune defenses. This is important for the development of novel therapies for atherosclerosis and could also help to underpin development of new approaches to combat pathogens with resistance to other treatments.

The LHC will restart data taking in May 2022, for its third Run. This provides a unique opportunity to do new measurements of B hadrons properties with the ATLAS experiment. Recent results from the LHCB experiment point to lepton flavour universality violation effects. If these effects are indeed confirmed, they would seem to point to new, beyond-standard-model physics. ATLAS is sensitive to such effects, and therefore will help to confirm or refute the discrepancies, and possibly point out which new physics is behind them. Genome instability is a hallmark of inherited human disorders and age-related diseases such as neurodegeneration and cancer. Eukaryotic genomes are replicated by complex molecular machines called replisomes, which frequently encounter naturally occurring ‘obstacles’ that can prevent faithful DNA replication and induce genome instability. Covalent DNA-protein crosslinks (DPCs) are one such obstacle and are estimated to occur at thousands of sites/genome/day in proliferating cells. Eukaryotic cells require multiple mechanisms to overcome DPCs, including replication-coupled proteolysis (‘DPC repair’). The objective of the project is to uncover the molecular mechanisms of replication coupled DPC repair. The project will involve in vitro reconstitution and biochemical characterisation of key steps in the eukaryotic DPC repair pathway, which will be imaged using time-resolved cryo-EM. The research will benefit from access to cutting-edge electron microscopy facilities at the Core Facility for Integrated Microscopy (CFIM; https://cfim.ku.dk/), which houses Copenhagen’s node of the Danish Cryo-EM Facility.

Faba bean is a protein crop with high yield potential under cold, northern climates. Its seeds accumulate high levels of protein, antioxidants, and micronutrients (e.g. iron & zinc), making it an ideal crop for inclusion in plant-based food systems in Scandinavia. However, faba bean seeds also accumulate high levels of condensed tannins and phytic acid, which are anti-nutrients that have a negative impact on protein digestibility and micronutrient bioavailability.
