1. Post-doctoral fellows for multiple NCI-funded projects to study the mechanisms of action of the MYC family of oncoprotein transcription factors
Summary of Postdoctoral Fellowship:
The Tansey laboratory has a long standing singular focus on MYC, one of the most important oncogenic drivers in human cancer. There is broad agreement that targeting MYC could form the basis of broadly effective anti-cancer therapies, yet MYC itself is undruggable. The premise of our work is that a detailed mechanistic understanding of MYC is the fuel that will propel MYC into the druggable universe. We use state of the art molecular, genetic, genomic, and proteomic approaches, together with sophisticated in vivo mouse models, to expose and validate novel and actionable vulnerabilities in the MYC network. Current research in the laboratory is divided into three areas, each devoted to a specific MYC cofactor—WDR5, HCF-1, and the SWI/SNF chromatin remodeler. Recent publications in eLife (PMID: 33416496), Cell Reports (PMID: 33472061), PNAS (PMID: 31767764), and Nature Communications (PMID: 31043611) illustrate our approaches and current project trajectories. More information about the laboratory environment and our current research efforts is available at tanseylab.com.

2. Postdoc position to study how 2nd messenger signaling molecules alter the structure of nuclear receptor transcription factors
Summary of Postdoctoral Fellowship:
We recently used integrated biophysical techniques to determine the unliganded, ground-state structure of a full-length nuclear receptor bound to DNA. We now want to add regulatory ligands to this ground state to determine how activating or repressing small molecules (drugs and physiological ligands) alter the structure of this protein. The techniques you’ll use will mainly be based on mass spectrometry (HDX and/or chemical X-linking). The ideal candidate will need to have an interest in helping others in the lab and have skills in structural biology, protein expression/purification, SAXS, X-ray crystallography and/or mass spectrometry-based proteomics.

3. Postdoc position to study how 2nd messenger signaling molecules generated in chromatin alter histone acetylation and gene expression
Summary of Postdoctoral Fellowship:
We recently discovered a new gene regulatory mechanism in human cell lines. This new mechanism revolves around a kinase our lab studies, which generates inositide 2nd messenger signaling molecules. We discovered this kinase is recruited to thousands of transcriptional start sites, regulating gene expression in a kinase-dependent manner and altering histone acetylation and methylation. The ideal candidate will need to have an interest in helping others in the lab and have skills in functional genomics. Experience with or eagerness to learn ATAC-seq, chromatin immunoprecipitation (ChIP),RNA-seq, RT-qPCR, qPCR and other gene expression analyses is ideal.

29. Postdoctoral position in novel biomaterial, designed for injection
Summary of Postdoctoral Fellowship:
We are working on a novel biomaterial, designed for injection in/adjacent to a treatable metastatic lesion and capable of recruiting and reprogramming tumor associated macrophages to enable local and systemic anti-cancer immunity. We hypothesize that local polarization of tumor-associated macrophages (TAMs) toward an M1 phenotype will create a metastasis microenvironment responsive to cancer immunotherapies. We are fabricating and characterizing TAM-attractive cryogels (TAC) to induce repolarization of macrophages. We characterize macrophage repolarization by TACs in vitro and in ex vivo human breast tumor explants. Additional studies use implanted TACs in a mouse model of breast cancer; we quantify immune responses and changes in tumor progression.
