Common mental disorders (CMD) are the leading causes of long-term sickness absence. You will explore the relation between shift work (especially night work), long working hours and CMDs, as well as the interaction with psychosocial work environment. The goal is to show that routinely collected data can be used to a) identify risk factors for CMD, b) identify workplaces with high risk of sickness absence and c) give the employer recommendations on how to design healthy schedules. We have established a cohort of over 60,000 individuals employed by Stockholm Region between 2008 and 2021. Detailed information on working hours is retrieved from registry-based payroll data for each working day during the study. You will use longitudinal multivariate analysis to investigate working hours in relation to CMD and sickness absence, also including interaction between working hours and psychosocial work environment. In a non-randomized pseudo trial design, we will study the effect of changing work schedules on CMD. Finally, we will use ensemble methods to identify models that can predict workplace risk of long-term sickness absence among employees. The employment is temporary for two years.

Our lab has invented the first method for producing artificial spider silk fibers and our current main research focus lies in improving the mechanical properties of these by protein engineering and development of the spinning device. The group is well funded by e.g. an ERC consolidator grant. By using next generation sequencing technologies we have increased our understanding of the silk production machinery and will use this knowledge to improve the artificial spider silk fibers further. We have published several articles in high ranked journals (Arndt et al. Adv Funct Mater. 2022; Rising & Johansson ACS Nano. 2021; Schmuck et al. Materials Today. 2021; Andersson et al. The Simon research group is studying regeneration processes in aquatic salamanders. Natural examples of regeneration, such as salamanders, may unravel novel strategies for cell replacement of damaged or lost tissues. We study regeneration mechanisms in an aquatic salamander, the newt, which has the widest repertoire of regenerative abilities among tetrapods. During the past decade, our laboratory has developed several methods, which has made these outstanding questions experimentally tangible. We place our findings on newt regeneration into an evolutionary context and test our results in a variety of mammalian in vivo and in vitro assays. The main target tissue of our investigations in the context of limb regeneration is skeletal muscle. Skeletal muscle can contribute to limb regeneration in two ways, either by activation of quiescent stem cells or by dedifferentiation of differentiated muscle fibers. By various cell tracking strategies, we try to compare these two processes to each other in terms of their relative contribution to the limb. We also characterize molecular processes involved in the generation of limb progenitors from skeletal muscle.

Our lab investigates stromal scar formation and its impact on axonal regeneration and functional recovery following spinal cord injury. We discovered a new population of perivascular cells, called type A pericytes, as the source of fibrotic scar tissue (Göritz et al., Science, 2011) and identified pericyte-derived scar formation as a therapeutic target to achieve axonal regeneration and functional recovery following spinal cord injury (Dias et al., Cell, 2018). We recently demonstrated that glutamine levels are lower in white adipocytes from obese compared to lean subjects and that this leads to white adipose tissue (WAT) inflammation (Petrus, Lecoutre et al, Cell Metabolism, 2020). Our new preliminary data in human and murine WAT, reveal that these changes are associated with decreased expression of Glutamine Synthase (GS, encoded by GLUL) and increased expression of glutaminase (GLS, encoded by GLS), the two enzymes governing glutamine metabolism. This suggests that glutamine turnover is a major determinant of adipocyte function. To further dissect the pathophysiological role of our observations in vivo, we have generated adipocyte-specific Gls and Glul knockout mice where we will study the effects of different perturbations on WAT and whole body metabolism. These analyses will be complemented by state-of-the-art mechanistic studies in human adipocytes in which we for instance will perform metabolic flux analyses following CRISPR/Cas9-mediated engineering of the glutamine pathway.
