The Center for Immuno-Oncology (CIO) functions as a multidisciplinary and interdisciplinary translational and clinical research programmatic effort with the goal of developing novel immunotherapies for cancer. The Fellowships in Surgical Oncology Research and Cancer Immunotherapy at the National Cancer Institute (NCI) encompass two time-honored programs combining bench-side research in surgical oncology with training in clinical research through direct patient care. However, the underlying mechanism is unknown. Recent progress in cancer immunotherapy has emphasized how multifactorial and systemic immune responses must be to trigger tumor eradication. The need for quantitative and actionable models is being acutely felt to expand our fundamental understanding of the immune system, and to usher new immunotherapies. The Altan-Bonnet group is studying the interplay between innate and adaptive immune responses across the whole organism. Signaling from Eph receptor tyrosine kinases and the ephrin transmembrane ligands affect morphogenesis during development and have been shown to play an instructive role in angiogenesis, as well as tumor cell invasion. Since we found a connection between the key Wnt signaling scaffold, Dishevelled, and ephrinB proteins, we have extended our analysis to examine how the intersection of Wnt/Planar Cell Polarity (PCP) and ephrinB signaling alters the mechanisms of signal transduction of these pathways. Moreover, we investigate how the output of these pathways lead to the creation of highly ordered intricate structures during development. Both Xenopus (Frog) and mammalian tissue culture systems are employed in projects that involve the functional and molecular characterization of the cellular and developmental effects mediated by the ephrinB and Wnt pathway proteins. A postdoctoral position is available in 2022 to study the role of ephrin and Wnt signaling pathways in cell movement and morphogenesis. Our long-term working goal is to explore the molecular and cellular biology of thoracic cancers, with special focus on those affecting surface cavities like mesothelioma and thymic epithelial tumors, in order to develop strategies that can improve diagnosis and treatment paradigms. Other tumors of interest include lung carcinomas or metastatic tumors to the thorax, neuroendocrine tumors, esophageal cancer etc. The scope of our projects are translational by design. Project I: design novel models of oncogenesis and perform comprehensive genomic and physiologic characterization that will facilitate developing novel, clinically relevant molecular biomarkers. Project II: investigate microRNA-mRNA interactions to inform on the pathogenic signaling pathways relevant to tumor initiation, invasion, metastasis, and resistance in malignant mesothelioma. With these projects, we have translational aims of novel therapeutic targets based on an understanding of the active cancer-specific mechanisms. Also, we are developing novel in vivo delivery platforms for nucleic-based agents that require pre-clinical testing in mouse tumor models. The culmination of these projects will be linked to clinical human testing in these thoracic cancers of interest.
