Summary of Doctoral Project:

The Kovar group tackles unresolved questions of pediatric solid tumor origin, pathogenesis and progression with a focus on bone sarcomas in order to identify new treatment options. In this project funded by the Austrian Science Fund FWF, we will establish innovative 3D models of solid tumors in their lung metastatic niche to recapitulate the full complexity of tumor/microenvironment interactions. These models shall be used in high-throughput pre-clinical studies to personalize drug selection for patient treatments. This project, will employ an array of state-of-the art technical approaches, including single-cell (sc)RNA-seq, spatial transcriptomics, organoids, and 3D-bioprinting.

2. PhD student (m/f/d) in Immune Cell Signaling

Summary of Doctoral Project:

Our group (https://www.ccri.at/eva-koenig-group/) studies the role of natural killer cells in tumor surveillance and aims to identify novel ways to enhance NK cell functions in the fight against cancer. The current position extends our long-standing interest to study the non-canonical role of STAT1 in the synapse of NK cells that is formed when they attack tumor cells. We use DNA barcoding technologies, multi-omics approaches (single-cell and bulk RNA-seq, ATAC-seq), genetic engineering (CRISPR/Cas9 gene knockout/-in, siRNA knock-down), mouse models and a diverse array of in vitro functional studies (including multi-color flow cytometry and confocal imaging). The current position is offered for four years and financed by the stand-alone grant P34832 of the Austrian Science Fund (FWF).

3. PhD Student (m/f/d) – drug resistance in pediatric sarcomas

Summary of Doctoral Project:

The Tomazou group (https://www.tomazoulab.org/) seeks to recruit a PhD student who will take on a highly ambitious research project aiming to establish novel mechanism(s) of drug resistance in pediatric tumors. Using ribosome footprinting, functional genetic screens (both in vivo and in vitro) and single cell technologies, we will test the hypothesis that a “ribosome code” (heterogeneity of ribosome composition resulting in selective mRNA translation) mechanistically contributes to drug resistance in pediatric sarcoma cells. This project is supported by the FWF 1000 ideas program that promotes innovative research with high scientific and transformative potential.

The aim of the project is to use the framework of Tinbergen’s four essential questions to understand the mechanism through which dietary intake and internal synthesis n-3 LC-PUFA affects biochemical and cellular quality of brain and cognitive traits of brown trout. The project puts especially strong emphasis on ecologically relevant context of the research. In one of three interlinked Post Doc and Ph.D. projects, we will focus on understanding the molecular basis of action of mutanofactins, as single compounds and their mixtures to assess potential synergistic effects. This includes analysis of effects on bacterial growth, stress tolerance and biofilm formation of consortia including competing oral streptococci and cohabiting cariogenic biofilm species. We will create a hierarchical model interlinking the interaction of mutanofactins to structural and mechanical properties of biofilms to understand the presumably novel mechanisms by which these secondary metabolites control cariogenic biofilm formation.
