3. Fully Funded PhD Position in Medical Science – NOX2-derived ROS in the tumor milieu

Summary of PhD Program:

This doctorate project aims at understanding how NOX2-derived reactive oxygen species (ROS), produced by intratumoral myeloid cells, shape the tumor microenvironment. NOX2 is an enzyme complex that is almost exclusively expressed by myeloid cells. Upon activation it generates ROS that contribute to host defense against microbes, but when released extracellularly, ROS exert and array of effects on neighboring cells. We have previously seen that NK and T cells cells are highly sensitive to the toxic actions of ROS and that NOX2 inhibition enhances the ability of cytotoxic cells to kill tumor cells. In addition, our preliminary studies show that the NOX2-derived ROS released within tumors also exert direct effects on cancer cells as they stimulate epithelial to mesenchymal transition (EMT) which enhances the metastatic capacity of the cancer cells.

Antibiotics and Antimicrobial Resistance (AMR) pollution in urban rivers receiving WWTP effluents is common, but their fate, and environmental risk remains poorly studied in river sediment. Context-dependency (ecological abiotic and biotic factors) influences the assembly of natural communities of bacteria and may affect their sensitivity to antibiotics and AMR. This PhD project will provide new knowledge on the interactions between pollutants (residues of antibiotics and AMR) and sediment bacteria, under altered abiotic and biotic contexts: a gradient of microbial diversity, pre-exposure history and/or pollution levels. Experimental studies using natural microbial communities in microcosms facilities and field sampling will be implemented for hypothesis testing.

7. Fully Funded PhD Position in Molecular Microbiology

Summary of PhD Program:

This project is part of the larger collaboration funded by JPIAMR called NewResGenes: Informing genomic surveillance by uncovering, phenotyping and prioritising resistance genes to new antibiotics. We will leverage variations that exist in natural populations of pathogenic bacteria to identify genomic variants underlying resistance to new antimicrobials. We have previously adopted a high-throughput system to screen growth kinetics and yield of Escherichia coli on agar plates capable of screening ~50,000 colonies in one experiment and we collected and sequenced >2000 diverse Escherichia coli strains isolated from a wide variety of environments. This includes strains isolated from the pre-antibiotic era and recent highly resistant clinical isolates.

The Morud Lekholm research group studies dynamical changes at ligand-gated ion channels using molecular biology, biochemical and biophysical techniques.
