We are looking for a scientist or engineer with a strong technical background in transmission electron microscopy, who is interested in performing research in methods and technology for cryo-electron microscopy and tomography. The Max Planck Institute of Biochemistry has a vibrant community of electron microscopists that develops and uses novel cryo-electron microscopy, tomography, and FIB-SEM methods and that works closely with industrial partners. The institute has a rich history of successful hardware and software development in this area and currently operates a large fleet of TFS electron microscopes including four Titan Krios, an Arctica, two Glacios, an Arctis plasma-FIB and two Aquilos FIB-SEMs.

2. Postdoctoral Job
Summary of Postdoctoral Fellowship:
A major focus of the Akhtar lab is understanding long noncoding RNAs and RNA remodeling by the RNA helicase DHX9. Our most recent work in Drosophila revealed that phase separation between the unstructured CTD of MSL2 and the roX RNAs is responsible for establishment of a gel-like condensate that promotes local transcriptional activation (Valsecchi et al., 2021 Nature). In the future the lab will dissect the fascinating question of what properties of this phase-separated compartment are responsible for guiding transcriptional upregulation at genomic loci found inside it. In mammals our lab has identified DHX9 as a key enzyme in protecting cells against the harmful effects of retrotransposon-derived secondary RNA structures (Aktas et al., 2017 Nature).

3. Postdoctoral Job
Summary of Postdoctoral Fellowship:
The Akhtar lab has a long-standing interest in gene dosage and understanding how epigenetic modifiers fine-tune transcription at both individual loci and at the chromosome-wide level (Valsecchi et al., 2021 Nature). While most genes tolerate mutation of one allele, dosage-sensitive genes known as haploinsufficient genes require the transcription of both maternal and paternal alleles. Mutation in just one allele of a haploinsufficient gene leads to disease. In our most recent work we utilized an elegant hybrid mouse model to reveal that the MSL complex component and chromatin regulator MSL2 ensures biallelic expression of this important class of genes (Sun et al., 2023 Nature).

The lab generates large quantities of deep sequencing datasets, including RNA-seq, ChIP-seq, ATAC-seq and Nanopore long read sequencing, which we analyse using a combination of adaptation of existing bioinformatic tools and development of new analysis tools. Members of the lab also contributed to the deepTools2 suite (Ramírez et al 2016 Nucleic Acids Research) and snakePipes package (Bhardwaj et al 2019 Bioinformatics) developed by the bioinformatics facility headed by Dr. Thomas Manke.

The current position focuses on exploring different ML and AI approaches to develop new ESM representations for key processes on land using Earth observation data. In particular, we are looking to improve biosphere-atmosphere interactions in terms of land carbon uptake and ecosystems’ response to climate extremes in the ICON land surface model from short to long time scales, looking at improving the simulation of ecosystem response to water and heat stress, phenological regimes and shifts in response to climatic changes, up to the dynamics of vegetation mortality and carbon turnover. Conceptually, we aim inducing observation-based ecosystem functioning through hybrid modelling approaches that combine different observational products.

The project aims to exploit a conceptually new approach to develop therapeutic antibodies targeting high-value drug targets like G-protein coupled receptors (GPCRs) or ion channels. It centers on creating and validating cellular assays for functional screenings of antibody candidates, including establishing systems for higher throughput antibody production. Additional projects may involve further antibody characterization, including binding kinetics (ELISA, Flow Cytometry, BLI), thermostability (DSF), aggregation propensity (SEC) or hydrophobicity (HIC).
