Our laboratory is part of the Neuroscience Department of the Institut Pasteur and gathers psychiatrists, neuroscientists and geneticists to understand the causes of autism. We previously identified the first mutations in the NLGN-NRXN-SHANK pathway, which has a key role in synapse formation within the brain. Our results highlight the genetic heterogeneity of autism, but also point at common biological mechanisms that could constitute relevant targets for new treatments. We are leading the genetic work package of AIMS-2-TRIALS, the largest European project on autism research. The project is focused on deep-phenotyping of individuals with autism and controls including brain imaging (EEG and MRI) and a battery of cognitive tests. Our group is currently developing new methods for analyzing whole genome and brain imaging data as well as new paradigms for characterizing mouse social and vocal behaviors.

4. Five Postdoctoral and Engineer positions – Epigenetic-based therapies to fight infectious diseases

Summary of Postdoctoral Fellowship:

Five positions (four postdoctoral and one engineer) are available to join an interdisciplinarity consortium, located in the Paris area, to study the interplay between epigenetic regulation and infectious diseases. Our objective is to find new therapeutic approaches to fight infection using epigenetic drugs. Our consortium combines expertise in the biology and epigenetics of infection (5 teams), in proteomics (1 team) and in drug-design and medicinal chemistry of epigenetics (2 teams). The partners are internationally-oriented research structures, located at the Pasteur Institute (6 laboratories), University of Paris (1 laboratory) and University Paris Saclay (1 laboratory). The strengths of the consortium are: (1) a strong track-record showing that pathogens modulate host epigenetic mechanisms; (2) considerable resources generated by medicinal chemists; (3) in-depth expertise on seven biological models, from in vitro and in cellulo to in vivo models. The research environment includes state-of-the-art equipment and facilities for the project. Salaries are funded for at least two years and will be adjusted according to the experience of the candidate.

In brief, invasive fungal infections (IFIs) are associated with high mortality rate. Despite antifungal drug treatment, the prognosis of IFIs is poor due to inadequate efficacy of the drugs, variable drug- susceptibility across fungal species due to their complex biological cycle and the emergence of drug-resistant strains. This demands new/alternative therapeutic strategies. On the other hand, cell wall is the fungal first component to interact with the host immune system. However, polysaccharides, the main constituents of the fungal cell wall are under-explored in the context of immunotherapy. This is due to several drawbacks associated with the fungal cell wall polysaccharides. 7. Postdoctoral position – Mechanisms of HIV control in the absence of antiretroviral treatment

A funded post-doctoral position is available to develop a research program in the context of the ERASE HIV collaboratory studying the immune mechanisms contributing to HIV remission. The successful applicants will develop their research in the “HIV Inflammation and Persistence Lab” at Institut Pasteur in Paris, under the supervision of Dr Asier Sáez-Cirión. Dr Sáez-Cirión’s group is interested in understanding the mechanisms underlying control of HIV infection in rare individuals who are able to maintain undetectable viral loads for very long periods of time in the absence of antiretroviral treatment. The research program will be focused on the processes that lead to the development of optimal immune responses to counteract infection naturally or after early treatment initiation. Experimental approaches will cover cellular biology, immunology and virology.

8. Post-Doctoral research fellow: Characterizing immune responses following combination therapies in HBV infections

Summary of Postdoctoral Fellowship:

Current treatment strategies for HBV are life-long and rarely curative. Although HBV infects hepatocytes, its evasion and modulation of the immune response is key to the pathophysiology. The goal is to analyze the response to HBV during and after treatment with a combination of antiviral and immune modulating therapies using an immune competent humanized mouse model. Innate and adaptive immunity will be probed to determine whether efficient immune responses may be restored, leading to seroconversion and a functional cure off treatment. We aim to decipher the immune response leading to viral persistence versus viral control and uncover predictive biomarkers of each state.

9. Post-doctoral position: Immune- brain communication mechanisms

Summary of Postdoctoral Fellowship:

The brain is very hard to cure. Drugs have a hard time crossing the blood-brain barrier and if they do – the side effects of their activity in such a fragile environment may be disastrous. Our lab strives to describe and uncover an alternative approach to brain treatment. We study how peripheral signals, such as immune cells present at the brain borders, shape brain physiology, how the dysfunction of such communication pathways can contribute to brain aging or disease, and how we can exploit such pathways to cure the brain. We especially explore the immune niche of the choroid plexus (one of the brain borders), and the roles and signatures of microglia and border associated macrophages (Baruch, Deczkowska et al., Science, 2014; Schwartz & Deczkowska, TrendsImmune, 2016; A Deczkowska, O Matcovitch-Natan et al, Nat.Comms. 2017; Deczkowska et al., Nat.Neurosci. 2018; Deczkowska et al., Cell. 2018, 2020). The work includes single cell RNA-seq approaches, state of the art tools of immunology and neuroscience (incl. behavioral tests), mouse models, lots of genomic data and more!

10. Postdoctoral position in Stromal Immunoregulation

Summary of Postdoctoral Fellowship:

One funded postdoctoral position is available in the lab “Stroma, Inflammation and Tissue Repair” (Institut Pasteur, Department of Immunology, co-affiliated with Developmental and Stem Cell Biology) to explore the crosstalk between stromal cells and immune system in the context of chronic inflammation, diet-microbiome-adipose tissue axis and the metabolic syndrome. The stromal microenvironment is increasingly recognized as an upstream organizer of tissue homeostasis and immune responses. Our lab has performed pioneering work on the role of stromal cells in tissue homeostasis and how perturbation of stromal pathways impacts on repair responses and inflammatory diseases (Dulauroy, Nature Medicine 2012; Stzepourginski, PNAS 2017; Di Carlo, JCI 2018; Benabid, Current opinion Immunol 2020; Jacob, Cell Stem Cell 2022). In this project, we want to understand how stromal cells sense their environment to maintain tissue homeostasis while ensuring efficient responses to external challenges, with a focus on the adipose tissue and mucosal tissues. To go beyond the state-of-the-art, we will perform phenotypic, functional and transcriptomic based screens (bulk/ single cells RNAseq) to identify stromal subsets and genes that regulate tissue homeostasis and remodeling, building on preliminary data obtained in the lab.
