Cyclic peptides are able to inhibit difficult targets such as protein-protein interactions and protein-RNA interactions with high potency. Although cyclization can improve cell permeability under specific conditions, most cyclic peptides are not able to pass cellular membrane effectively. To make cyclic peptides more widely applicable as chemical biology tools and drug candidates, strategies to overcome this barrier are required. Cyclic disulfides have been demonstrated to be able to undergo disulfide exchange with cell surface receptor thiols and by doing so mediate cell uptake of various cargoes. In this project you will focus on the synthesis of cyclic disulfide amino acids that will simultaneously facilitate peptide cyclization as well as provide a handle for cell permeation via the disulfide exchange pathway. The peptides will then be evaluated using cell biology and microscopy techniques.
