The lab is developing new, general approaches to control the site and timing of enzyme activity. Enzymes have great potential as therapeutic and diagnostic agents due to their high specificity and catalytic activity, but the ability to better control the timing and location of enzyme function would enable the use of a wider variety of enzymes for therapeutic purposes. Our approach for enzyme programming relies on attaching an enzyme to its small-molecule inhibitor through an allosteric DNA tether. Upon interaction of the DNA tether with its complement strand, the ensuing conformational change activates the enzyme by dislodging the inhibitor from its active site. We have established the utility of DNA-programmed intrasterically-regulated enzymes in developing constructs for fast, sensitive, and PCR-independent isothermal detection of label-free nucleic acid sequences; carrying out complex logic operations; programming the activity of streptokinase, a clinically-used fibrinogen activator; and specifically killing targeted cancer cells.
