The University of Pennsylvania Department of Ophthalmology is seeking a Postdoctoral Fellow for research on the pathobiology and genetic risk factors of primary open-angle glaucoma (POAG). The successful candidate will participate in a large NIH-funded study uniquely focused on identifying phenotypic and genetic variables associated with POAG in African Americans. POAG is the leading cause of irreversible blindness in the world today. This heterogeneous disease is known to manifest differently across and between genetic populations, yet most POAG research has focused exclusively on genes of European ancestry. POAG is five times more common and ten times more likely to cause blindness in African Americans compared to European Americans, and occurs earlier with more rapid progression in this over-affected population. Through the Primary Open-Angle African American Glaucoma Genetics (POAAGG) study, we have collected DNA of over 10200 African Ancestry cases and controls in Philadelphia with over 90% of phenotypic and demographic information for cases. All cases and controls were recruited in Philadelphia and we have the participants approval to re- contact them for future studies. In analyzing this data, we aim to develop better-targeted treatment and diagnostic approaches for POAG, a devastating disease affecting many of our patients.

4. Postdoctoral Research Associate in Stem Cell Biology

We are seeking a highly motivated individual for a postdoctoral position in the Gadue laboratory in the Center of Cellular and Molecular Therapeutics at the Children’s Hospital of Philadelphia. The Gadue laboratory focuses on using human ES and iPS cell models to study human pancreatic development and diabetes. The NIH funded position aims to study multiple forms of diabetes using genome engineered stem cell lines with a focus on endocrine lineage cells. Projects available include using the stem cell model to functionally validate variants discovered by genome wide association studies and their target genes in beta cell biology, especially in regard to cellular stress and how this relates to diabetes pathogenesis. In addition, projects on the study of monogenic forms of diabetes are also available, focusing on HNF1A and its downstream targets in endocrine cell development and function. Techniques used widely in the laboratory include CRISPR based genome engineering, stem cell culture, in vitro differentiation, flow cytometry, fluorescent microscopy and various genomic techniques (RNA-seq, ATAC-seq etc…).
